Validation Data

Figure 1. Detection of HLA-A2 expression in spleen and intestine by RT-PCR.

Wild type: only one band at 400 bp with primers F1/R1 (mH2-D1), Heterozygous: one band at 400 bp with primers F1/R1 (mH2-D1) and one band at 664 bp with primers F2/R2 (hHLA-A2), Abbr.. M, DNA marker, HO, homozygous, HE, heterozygous, WT, wild type.

The above results were in line with expectations. Wild type mice showed high expression of H-2Db, while HLA-A2 humanized heterozygous mice showed high expression of H-2Db and hHLA-A2. There was no significant difference in the proportion of various immune cells to CD45 positive cells between wild type mice and HLA-A2 humanized mice, indicating that the genetic modification had no significant effect on the proportion of immune cells in mice.
